L-丙氨酸（L-Ala）作为重要蛋白质的构建块起着至关重要的作用。 L-丙氨酸主要由来自乳酸的肌肉细胞合成并通过肝脏吸收到血液中。 它被谷氨酸丙 酮酸转氨酶转化为丙 酮酸，进入代谢主流。 L-Ala对葡萄糖的产生和血糖管理至关重要，并在免疫系统和预防肾结石方面发挥重要作用。 L-丙氨酸的缺乏通常是营养不良，低蛋白饮食和压力的标志。 AAT Bioquest的Amplite 荧光L-丙氨酸测定试剂盒提供了一种灵敏的荧光测定方法，用于量化生物样品中的L-丙氨酸。 它利用一种酶偶合反应释放过氧化氢，Quest Fluor L-丙氨酸传感器在Ex / Em = 540/590 nm处使用荧光酶标仪进行检测。检测限：~3uM，任何带有-SH基团的物质，如DTT、半胱氨酸，都会干扰测定。

样品实验方案
简要概述
1.准备测试样品以及连续稀释的L-丙氨酸标准品（50 µL）
2.加入等体积的L-丙氨酸工作溶液（50 µL）
3.在37°C下孵育30分钟至1小时
4.监测Ex / Em = 540/590 nm的荧光强度
溶液制备
1.储备溶液
所有未使用的储备溶液应分为一次性使用的等分试样，并在制备后储存在-20°C下。 避免重复冻融循环。
1.1 Quest Fluor L-丙氨酸传感器储备溶液（200X）：
将55 µL DMSO（组分E）添加到Quest Fluor L-丙氨酸传感器（组分A）中，制成200X Quest Fluor L-丙氨酸传感器储备溶液。
1.2 L-丙氨酸标准溶液（1 mM）：
将10 µL 100 mM L-丙氨酸（组分D）加到990 µL PBS（pH 7.0）中，得到1 mM L-丙氨酸溶液。
2.标准溶液
L-丙氨酸标准
向900 µL PBS中加入100 µL 1 mM L-丙氨酸标准溶液，制成100 µM L-丙氨酸溶液（AS7）。进行1：2连续稀释以获得连续稀释的L-丙氨酸标准品（AS6-AS1）。
3.工作溶液
3.1 将5 mL测定缓冲液（组分C）加入一瓶酶混合1瓶（组分B1）中，并充分混合。
3.2 将100μLddH2O加入一个酶混合物2小瓶（组分B2）中并充分混合。
3.3 将整个小瓶（100μL）的Enzyme Mix 2和25 uL的200X L-丙氨酸传感器储备溶液转移到Enzyme Mix 1瓶中； 混合均匀。 注意：工作解决方案不稳定。 立即使用，避免直接暴露在光线下。

1.根据表1和表2提供的布局，准备L-丙氨酸标准品（AS），空白对照（BL）和测试样品（TS）。对于384孔板，每孔使用25 µL试剂代替50 µL。
2.将50 µL L-丙氨酸工作溶液添加到L-Alanine标准品，空白对照和测试样品的每个孔中，以使L-Alanine测定的总体积为100 µL /孔。 对于384孔板，将25 µL L-丙氨酸工作溶液添加到每个孔中，总体积为50 µL /孔。 注意：在pH 6.5至7.0下运行L-丙氨酸测定。
3.将反应混合物在37°C孵育30分钟至1小时。
4.使用荧光板读数器在Ex / Em = 540/590 nm（截止：570 nm）下监测荧光的增加。
