KU812E is a clone of KU812 (ATCC CRL-2099). The parent KU812 cell line was established from the peripheral blood of a patient in blast crisis of chronic myelogenous leukemia. The cells contain at least one Ph1 (Philadelphia) chromosome. The cell line has some characteristics of basophilic leukocytes (Fc receptors, basophilic granules, histamine production), and are negative for lymphoid markers [PubMed: 85212346].
ATCC complete growth medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 20%. Atmosphere: air, 95%; carbon dioxide (CO2), 5% Temperature: 37.0°C
Subculturing:
Protocol: Cultures can be maintained by addition of fresh medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 1 X 10(5) viable cells/ml. Do not allow the cell concentration to exceed 3 x 10(6) cells/ml. Medium Renewal: every two to 3 days
