我们的滴度保证仅适用于包装进病毒的区域(从5’ ITR 到3’ ITR)小于腺病毒承载限度(38.7 kb,~野生型HAd5基因组长度36 kb的108%)的载体。超过该范围,超出的那部分序列在包入衣壳时会被剪切掉,导致包装出来的腺病毒颗粒含有不完整的genome。此外,对于如下载体,我们同样不能保证滴度:
- 包含对包装过程有不利影响的序列,例如毒性基因(如促凋亡基因);破坏包装细胞或病毒的完整性(如导致细胞聚集的膜蛋白)的基因 ;易于重排或者产生二级结构的序列(如重复序列或者高GC含量序列)。
- ⽤户提供的载体,因为我们无法控制载体质量。
我们提供第一代腺病毒(E1/E3-deleted Ad5)和嵌合型腺病毒(Chimeric adenovirus)包装服务。Ad5病毒的感染是通过病毒颗粒表面的纤突(Fiber)与细胞表面受体CAR(Coxsackie virus and adenovirus receptor)特异性结合而介导的。 并不是所有细胞都高表达CAR,Ad5病毒在某些不表达或者低表达CAR的细胞种类上的应用受限。我们开发了嵌合型腺病毒,它的感染是通过嵌合型纤突(Chimeric fiber)与细胞表面分子CD46特异性结合而介导的。

对照病毒
为了更好地开展您的实验,我们按照“对照病毒与定制病毒的生物学应用相匹配”的原则给您赠送或者推荐对照病毒。例如,如果定制病毒表达一个基因,则推荐的对照病毒表达荧光蛋白EGFP。如果定制病毒表达打靶某个基因的shRNA,则对照病毒表达一个Scramble shRNA。推荐的对照病毒信息如下表所示:
辅助病毒
为了更好地开展您的实验,我们按照“辅助病毒与定制病毒的生物学应用相匹配”的原则给您推荐辅助病毒。例如,如果定制化的病毒是一个gRNA腺病毒(只表达gRNA),则需要一个表达Cas9的辅助病毒,才能进行CRISPR编辑实验。推荐的辅助病毒信息如下表所示:

We ordered adenovirus from VectorBuilder to overexpress our protein of interest in primary mouse hepatocytes. The ordering process, including vector design, was very fast and easy. We received the plasmid and adenovirus within the time frame that was stated during the ordering process and the transduction efficiency of the adenovirus was very high, enabling us to use a low MOI. As a bonus, we also received the control Ad-EGFP adenovirus for free. In the future, I will not use any more siRNA or classical transfection methods on primary mouse hepatocytes, just adenoviruses from VB! 目前构建重组腺病毒系统的策略,主要有三种1)体外直接连接(clontech的ADENO-X系统)2)在细菌中重组的策略(adeasy系统)3)在293细胞中重组的策略(AdMax系统)国家人类基因组北方研究中心通过与美国Vector Biolab公司合作开发出了一套不同于以上策略的新的重组腺病毒系统。通过这套系统,实现了重组腺病毒载体构建,包装的高通量化。在不到一年的时间里,成功构建,包装了500多个腺病毒。通过对穿梭载体,骨架载体,包装细胞系的改进,提高了病毒的滴度,比目前其它腺病毒系统得到
