goodRIPA裂解液(RIPA的本意是Radio Immunoprecipitation Assay)是一种传统的细胞组织快速裂解液,主要用于从动物细胞和组织中提取可溶性蛋白,其裂解得到的蛋白样品可以用于常规的Western Blot、IP及Elisa等实验。RIPA裂解液的配方有很多种,根据其裂解强度大致可以分为强、中、弱三类。用RIPA裂解液裂解得到的蛋白样品,可以使用 BCA蛋白定量试剂盒 (货号:ZJ101或ZJ102)测定蛋白浓度。

多篇 SCI 发现用 RIPA 提蛋白存在问题,我们该如何应对?
:(1) 使用 RIPA 裂解液提取蛋白做 WB 会人为的增强或降低某些信号强度。RIPA 裂解液提取蛋白已经被证实有 10-30% 的蛋白会丢失在不可溶组分中 [12]。(2) 如果一个细胞 / 组织样品中某些特定的蛋白质可以在可溶组分和不可溶组分间转变,或者像上面所述受激活状态影响 [12] 的情况下,数据的解释可能成为一个大问题。(3) RIPA 裂解液提取的蛋白不适合用于目标蛋白的定性和 / 或定量分析。理想的总蛋白提取应该是什么样的呢?(1) 获取给定样品中完整的蛋白质图谱,要真实的反映所有蛋白
我的实验:将400ul的matrigel胶注入小鼠皮下形成的那么一个黄豆大的像凝胶一样的东西,会有血管长入。然后活体动物注射荧光素标记的Isolectin,可与新生血管内皮细胞结合,然后取出组织,溶解在RIPA裂解液中,匀浆后离心取上清再测定荧光强度。 该操作原文献只说用RIPA裂解液,没有给出成分。我用“RIPA”这个词检索,从一篇无关的英语文献中找到了配方。国内再查RPIA,共查到3-4种,大体成分均与英文文献相似,互相差1、2种成分或个别成分用量有一点差异,国内文献都是用来做
