In situ hybridization (ISH) using RNA probes is a valuable technique to characterize gene expression patterns in animal tissues. It provides valuable spatial information about gene expression. Compared to the nonradioactive alternatives,35 S radioactive ISH generally provides higher sensitivity. Here, we describe the procedure for35 S ISH on paraffin sections from the skeletal tissues of mouse embryos.
