一步定向克隆试剂盒是一款简单、快速、高效的多片段DNA定向克隆产品,可以不依赖于连接酶,不受载体和目的片段的酶切位点限制,直接用同源重组的方法将PCR产物定向克隆至任意载体的任意位点,完成目的片段与载体相连接的基因克隆技术。该技术操作简单,PCR产物一步定向克隆,目的片段与载体无缝连接,反应时间短至10分钟,阳性率达到95%以上,让您轻松完成实验。

应用

图例:在pUC19的EcoR I和Sal I酶切位点间插入一段基因,长度为800bp。 根据引物设计方案,设计800bp的正向扩增引物和反向扩增引物,如下所示: 正向引物: 5'-AAA ACG ACG GCC AGT –目的基因特异序列-3' 反向引物: 5'-CTT GCA TGC CTG CAG –目的基因特异序列-3' 将1μg环状pUC19质粒加入到70μl的酶切反应体系中,37℃消化过夜,内切酶使用量为EcoR I和Sal I各加入1μl,酶切完成后置于65℃加热20min,失活内切酶,并进行纯化回收。 使用Novoprotein Fast Pfu DNA聚合酶(货号:E031),对目的基因进行扩增并胶回收。 配置克隆体系反应后进行转化涂板和PCR鉴定。
