    Comments: Clone 15 was established in 1984 from a clone of HL-60 (ATCC CCL-240 ) that had been grown at an elevated pH (pH 7.6 to 7.8) for 2 months.
    This clone, picked from soft agar, was selected for its ability to undergo eosinophilic differentiation when treated with butyric acid.
    Initially, 80% of the treated cells will differentiate as eosinophils.
    However, with continued passage the line will gradually exhibit a modest degree of reversion such that a higher percentage of the cells will exhibit neutrophilic differentiation.
    The cells express IL-5 receptors and eosinophil granule proteins, and exhibit chemotaxis to eosinophil specific eosinophilotactins.
    Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.

    Subculturing: Protocol: Cultures can be maintained by addition or replacement of medium. Start new cultures at 2 X 10(5) viable cells/ml and subculture at 1 X 10(6) cells/ml.
    Medium Renewal: Every 2 to 3 days
    Preservation: Freeze medium: Complete growth medium supplemented with 5% (v/v) DMSO
