· Klenow说明
本Klenow是大肠杆菌DNA聚合酶I 经基因工程改造的产物。由于基因重组,其不含有DNA聚合酶I的5’→3’外切核酸酶活性,只具有5’→3’的DNA聚合酶活性和3’→5’的外切核酸酶活性。其中3’→5’的外切核酸酶的活性较弱,不适用于3’突出端的切平。
· Klenow用途
双脱氧法DNA序列测定(Sanger法)。去除PCR产物中残留的dNTP和焦磷酸盐
5’突出端的补平(参见SinoBio 实验方案)。
使用随机引物进行DNA标记。
· 质量保证
本Klenow经多次柱纯化,SDS-PAGE胶检测仅可见清晰单一的目的条带;PCR方法检测无大肠杆菌DNA残留,无核酸内、外切酶污染。
