      描述   脂筏是富含高水平胆固醇和鞘磷脂的微质膜结构域。脂筏涉及许多细胞过程,包括但不限于信号转导,膜运输和蛋白质分选。脂质修饰的蛋白质和一些跨膜蛋白质集中在脂筏中,而其他蛋白质被排除在外。脂筏还与质膜上的Na+ / K+ ATP酶相关。传统的脂筏分离方法使用蔗糖密度法或OptiPrep密度法,采用超高速离心法分离,需要大量的起始样品,而且该方法繁琐、耗时。为了克服传统方法的缺点,我们利用离心管柱技术开发了这款简单、快速的脂筏分离试剂盒。首先将样品的总膜组分分离,然后用含有非离子表面活性剂的缓冲液处理,最后使用台式离心机离心分离耐表面活性剂组分。整个过程无需使用密度梯度和超高速离心,可以在90分钟内从培养的细胞/组织中高度富集脂筏。

    试剂盒组分(20T):
    1. 缓冲液 A 15ml
    2. 缓冲液 B 10ml
    3. 缓冲液 C 10ml
    4. 研磨棒 2 个
    5. 离心管柱 20 个
    6. 收集管 20 个

     

    重要产品信息:
    1. 离心机请调整成 RCF 模式,所有离心步骤都需要在 4℃室温下或者低温离心机中进行。

    2. 研究蛋白磷酸化,磷酸酶抑制剂应在使用前加入缓冲液 A 中。如果担心蛋白降解问题可以分别在缓冲液A 和 B 中添加蛋白酶抑制剂。(请按照蛋白酶或磷酸酶抑制剂母液比例添加,例如母液是 100x,添加时
    按照 1:100 添加,即 1ml 缓冲液中添加 10ul 抑制剂)。

    3. 推荐使用 BCA 方法测定蛋白浓度。

    4. 研磨方式请按说明书操作,请勿使用液氮或匀浆仪等方式研磨。
