Use Glutamate Dehydrogenase in diagnostic tests for the determination of
ammonia, urea, L-glutamate, glutamate pyruvate transaminase and leucine
aminopeptidase.

Properties
Nomenclature: L-glutamate:NAD(P) + oxidoreductase (deaminating)
Molecular weight: ~2 200 kD for the associated enzyme with 8 subunits; 280
kD for one subunit.
Michaelis constants (Tris buffer, pH 8.0, +23°C):
L-glutamate: 1.8 x 10 -3 mol/l
NADP: 4.7 x 10 -5 mol/l
α-ketoglutarate: 7.0 x 10 -4 mol/l
NH 4 + : 3.2 x 10 -3 mol/l
NADPH: 2.6 x 10 -5 mol/l
Km values for NAD or NADH are difficult to obtain due to their inhibitory ac-
tion.
Inhibitors: 4-chloromercuribenzoate, Na 2 S, diethyldithiocarbamate,
1,10-phenanthroline,
8-hydroxyquinoline, NaN 3 , thyroxine, heparin, sulfonylcarbamides, Cu 2+ , Hg 2+ ,
Ag 2+ , Fe 3+ , Zn 2+ , K + , PO 4 2- , NO 3 -
Activators: Thioglycolic acid, b-mercaptoethylamine, EDTA, α, α'-dipyridyl
pH optimum: 8.0 (see figure)

pH stability: 5.5-6.5 (see figure)
Thermal stability: Up to +60°C (see figure)
Specificity: The oxidation of L-glutamate is stimulated by ADP and inhibited
by GTP. In contrast, the oxidation of alanine, leucine, isoleucine, methionine,
valine, norleucine, norvaline and 2-aminobutyrates is stimulated by GTP and
inhibited by ADP.
Remarks:
- Glutamate dehydrogenase suspension or solution can be dialyzed against
phosphate buffer, 10 mmol/l. Glutamate dehydrogenase molecules have
the tendency to associate in some test formulations, modified Glutamate
dehydrogenase minimizes this effect.

一种催化使氨还原性地固定到α -酮戊二酸上形成谷氨酸反应的酶。 EC. 1. 1. 1. 3[NAD( P) ] 1. 1. 1. 4( NADP)。 α -酮戊二酸 NH3 NADH H 谷氨酸 NAD 它广泛分布于生物中,在动物,存在于线粒体中。虽然反应是可逆的,但平衡具有朝向合成谷氨酸的倾向。因物种的不同,辅酶的特异性有所不同,在植物中是 NAD,酵母是 NADP,动物中虽不论那种辅酶都有作用。但 NAD则较其他辅酶的效力高出数倍。反应
C10 H17 N3 SO6 。为γ -L-谷氨酰 -L-半胱氨酞甘氨酸( 1)。谷胱甘肽如( 2)所示在活体内参与氧化还原反应。所以具有保护如组织蛋白酶、番木瓜酶及琥珀酸脱氢酶那样具有 SH基酶的 SH基作用。另外是催化甲基乙二醛形成乳酸的乳酰谷胱甘肽裂解酶( EC. 4. 4. 1. 5)不可缺少的辅助基质。在解毒作用中形成硫醚氨酸,被认为有毒物质先与谷胱甘肽结合使谷氨酸与甘氨酸部分切断。
反应。在缺乏氨基酸氧化酶的高等动物中,首先进行转氨酶所催化的反应(Ⅰ),再以谷氨酸为媒介,在谷氨酸脱氢酶催化的反应(Ⅱ)中生成氨,在进行氨基酸氧化脱氨的同时,通过逆反应参与氨基酸的生物合成。也有以丙氨酸为氨基供体的转氨酶。(2)谷氨酸、天冬氨酸等的氨基酸的酰胺基也能直接作为氨基供体,但这时被转移的是α-氨基,而酰胺基则作为氨波游离出来。(3)在动物的肝脏、微生物中发现鸟氨酸、r-氨基丁酸、β-丙氨酸等的。ω-氨基转移到α-酮酸的反应,在这种情况下,除α-酮酸外,醛类也能成为氨基受体。鸟氨酸特别
