本试剂盒适于检测哺乳动物细胞中的线粒体跨膜电势差(MMP)，并可用于在酶标仪上筛选凋亡抑制剂及激活剂等。线粒体膜电位试剂盒已用于测量线粒体膜电位。
Biochem/physiol Actions
该试剂盒利用JC-10，一种优于JC-1的替代品，用于测定细胞中MMP的损失。尽管JC-1在许多实验室中被广泛使用，但是当其在较高浓度下使用时，其水溶性差通常会导致水性缓冲液沉淀。在较高浓度下，JC-10表现出比JC-1更高的水溶性。与JC-1类似，JC-10是一种阳离子亲脂性染料，其浓缩并在具有极化线粒体膜的细胞线粒体中形成可逆的红色荧光JC-10聚集体(λex=540/λem=590nm)。在凋亡细胞中，MMP崩溃导致JC-10无法保留在线粒体中，并使染料返回到单体绿色荧光形式(λex=490/λem=525nm)。该试剂盒可用于监测细胞凋亡和筛选凋亡抑制剂和激活剂。
General description
由于电子传递链上酶的活动，线粒体在其膜上产生电位。在细胞凋亡过程中，线粒体膜电位(MMP)的崩溃与线粒体通透性转换孔的开放一致，导致细胞色素c释放到细胞质中，进而引发细胞凋亡级联反应的其他下游事件。
