【产品简介】
该产品彩包含从10 kDa到180 kDa共10种高度纯化并预染的重组蛋白质（10 kDa，17 kDa，25 kDa，33 kDa，43 kDa，55 kDa，70 kDa，95 kDa，130 kDa，180 kDa，均带His标签），其中70 kDa条带为橙色，10 kDa为绿色。表观分子量经过Thermo26610和26614非预染蛋白质分子量标准标定。
蛋白条带经彩色预染，便于实时观察电泳进程。
【产品应用】
作为SDS-PAGE和Western Blot实验中蛋白分子量大小的参照。
【产品组成及标准】
2×250 µL，-25~-15 ℃条件下储存 1 年，2~8℃可放置 2 个月，干冰运输。
【操作流程】
将本产品于室温下彻底解冻后轻轻摇匀，取5 µL（5×1.5 mm胶孔）上样，与样品同时进行PAGE电泳。预染蛋白在不同的缓冲体系下有不同的表观分子量，如果在该缓冲体系中事先用非预染蛋白质标定，可以大致确定蛋白质分子量。如下图，即为两种缓冲体系下的电泳示意图：
【注意事项】
本产品已包含1×SDS-PAGE上样缓冲液，可直接使用，请勿煮沸、稀释或加入还原剂等。
在低浓度凝胶中，多个低分子量蛋白可能同时泳动至凝胶前缘，请根据实际需要选择合适的胶浓度。
初次使用本产品时，建议进行上样量梯度预实验，确定合适的上样量，该操作可节约成本，同时获得更佳的实验结果。
在考染及His标签Western Blot实验中，建议上样量减少至需要量的1/5~1/3。
【问题讨论】
Q1：蛋白电泳时，该选择非预染Marker还是预染Marker？
A1：蛋白Marker是用来标示目标蛋白大小的，蛋白Marker可分为非预染蛋白Marker和预染蛋白Marker两种。
非预染蛋白Marker，没有附带染料分子或标记的蛋白Marker，只能在电泳完成后经过染色才能观察到，用于精确测定目标蛋白分子量。
预染蛋白Marker，与染料共价偶联的蛋白Marker，在电泳过程中就能实时看到Marker的电泳情况。用于监测电泳进行进度和情况、分子量的近似测定、估计蛋白质印迹（Western blot）过程中蛋白质转膜效率。预染Marker又分为单色预染和多彩色预染，Marker条带越多参考价值越大，同时也越难区分，若用不同颜色则容易辨认；单色的Marker通常利用其中某些条带加倍浓度来提示其大小。
˙Q2：使用预染蛋白Marker有哪些注意事项？
A2：
1）预染的蛋白Marker与染料的偶联作用导致在不同缓冲液中Marker迁移率不同，使用前需注意选择适当缓冲体系。若使用说明书中未涉及的电泳缓冲液，建议用非预染蛋白Marker对该预染蛋白Marker先进行分子量标定；2）在低浓度胶中，低分子量蛋白会泳动于染料前缘；
3）大分子量蛋白Western blot时需延长转膜时间或加高转膜电压。
