birkin 我在尝试转染某种细胞,载体是购买的商业载体(pCMV6),之前转染HEK293作为预实验,已经用WB证明有效,但是换到现在这种细胞之后就很奇怪,一开始想用短期转,结果WB做不出来,觉得可能是转染率太低,于是又改做稳定转染,用了G418筛选。花了近2个月,细胞稳定下来了,取样做RT-PCR,有相当强的mRNA表达(-RT和对照组转染也做了的,基本没有带,所以不会是污染),谁知做WB还是一点东西都没有……我都快疯了,既然能抗G418,又有大量mRNA
从Yeast Two-Hybrid中筛出一个蛋白,体外实验很成功,但在293中表达不成功. 已经尝试了多种载体,包括pCMV-tag 3B,pcDNA3,pcDNA3.1,检测不到蛋白的表达.钙转和脂质体都用过. 虚心求教!
