    RNA interference (RNAi) is a biological process in which RNA molecules inhibit gene expression, typically by causing the destruction of specific mRNA molecules. It is a valuable research tool because synthetic dsRNA introduced into cells can selectively and robustly induce suppression of specific genes of interest. RNAi may be used for large-scale screens that systematically shut down each gene in the cell, which can help to identify the components necessary for a particular cellular process or an event such as cell division. The pathway is also used as a practical tool in biotechnology, medicine and insecticides.

    shRNA is an advantageous mediator of RNAi in that it has a relatively low rate of degradation and turnover. Creative Biogene offers the generation of cell lines stably expressing shRNAs targeting a gene of interest. We will design your shRNAs and integrate them into virtually any cell line you choose.

    • Construct generation
    Synthesis and cloning of shRNA constructs targeting gene of interest into a suitable mammalian expression vector
    or a lentiviral vector followed by producing high titer lentivirus. Based on your requirements we choose
    the best strategy and corresponding vector system.
    • Stable cell clones selection
    Perform transfection or transduction on target cells and select stable cell clones.
    • Validation
    Validation RNAi gene silencing by real-time qRT-PCR and/or Western blot.
