目前在microRNA研究领域,microRNA生物学功能的阐明及microRNA特异性表达的机制研究仍然有很大困难。而microRNA靶基因PCR芯片可用来研究目的microRNA的功能。每款靶基因PCR芯片可以检测某一microRNA的84个经过实验验证或生物信息学预测的靶基因表达情况。一个microRNA可以调控一个或多个转录本,同样一个转录本可被一个或多个microRNA调控。靶基因PCR芯片所研究基因的选择是基于microRNA功能研究实验或生物信息学预测,所以此芯片可以在最大程度上验证或确定与microRNA表达模式相反的那些靶基因。

文献和实验、 miRNA靶基因的验证与miRNA靶基因 的预测方法相比,对miRNA靶基因进行实验验证的方法并不多,目前还没有一个快速、简便、高通量的鉴定方法。最直接的鉴定方法是, 利用荧光定量PCR及Westernblot方法分别检测转染或敲低miRNA后细胞中mRNA水平及蛋白水平的变化,从而确定miRNA与靶基因的对应关系。这种方法能够直接鉴定出miRNA的靶基因, 准确度高但不能鉴定miRNA的靶位点。转染或敲低miRNA:方式主要有两种,一种是构建miRNA过表达载体的稳定表达系统,一种是人工合成miRNA

【求助】miRNA mimic qRT-PCR检测可以提高多少倍?
nvonne 我购买的miRNA mimic 带FAM标记,本打算用流式检测转染效率,但发现荧光信号很弱,转染后采用定量PCR检测miRNA的表达变化,相对定量分析,发现mimic组比NC阴性对照组的miRNA表达量提高了100多倍,这虽然表明转染mimic成功,但总感觉这么高的变化有点太离谱了是不是?也没查到相关文献,谁能给我一些参考?非常感谢 zhujoker 顺转就是这么高,我的也是这样,有的能够上调1000倍
