Background:
Mucin 1, known as MUC1 in humans and Muc1 in other mammals, is a multifunctional protein expressed on the apical surface of most epithelial cells. The numerous biological activities attributed to MUC1/Muc1 in promoting cell growth and survival, are sometimes overshadowed by its name and classification as a tethered mucin. Muc1 is protective in the kidney in a mouse model of ischemia–reperfusion injury (IRI) by transactivation of the hypoxia-inducible factor 1 (HIF1) and b-catenin protective pathways. MUC1 does have an exceptionally strong glycan-dependent apical targeting signal that can redirect a basolaterally expressed protein to the apical cell surface in polarized epithelia; and the rate of MUC1 recycling from endosome to the cell surface is quite efficient and significantly higher than its rate of endocytosis (four-fold).
