| Background | The Src family of protein tyrosine kinases, which includes Src, Lyn, Fyn, Yes, Lck, Blk, and Hck, are important in the regulation of growth and differentiation of eukaryotic cells (1). Src activity is regulated by tyrosine phosphorylation at two sites, but with opposing effects. While phosphorylation at Tyr416 in the activation loop of the kinase domain upregulates enzyme activity, phosphorylation at Tyr527 in the carboxy-terminal tail by Csk renders the enzyme less active (2). |

文献和实验src 的产物 p60src 具有 TPK （酪氨酸蛋白激酶）活性，分子量约为 60KDa ，与细胞的增殖和转化有关。 Src 蛋白有四个结构域： （1） N 端十四烷基化位点是和膜结合的位点； （2） SH2 （ src homology region-2 ）结构域能和自身的或其它蛋白的磷酸化酪氨酸位点结合，传递信号。 （3） 416 位 Tyr 的自身磷酸化和细胞的转化活性有关。

即鸡肉瘤病毒(SRV)基因组中的基因，可使鸡产生肉瘤。是第一个鉴定的病毒癌基因。1970s,Peter Vogt分离到一种Rous 病毒的突变体，该突变病毒能够感染细胞并进行复制，但是不能致癌。由于该突变体只是丧失了将正常细胞转化为癌细胞的能力，因此推测突变的基因是诱导癌变的基因。后来的分析鉴定发现，该突变体只是缺失了一个基因。由于该基因的缺失，不能诱导肉瘤(sarcoma)的形成，故将此基因命名为src基因。
