1. Nature: AlphaFold 预测 98.5% 的人类蛋白结构

2021 年 7 月 22 日，英国 DeepMind 团队在 Nature 杂志发表了重磅文章 Highly accurate protein structure prediction for the human proteome。

该文章描述了 AlphaFold 对人类蛋白质组（人类基因组编码的所有蛋白质的集合）的准确结构预测，能对人类蛋白质组 58% 的氨基酸的结构位置给出可信预测。大规模的准确蛋白质结构预测，不得不说对结构生物学专业是项巨大的冲击与考验！

2. Nature Biotechnology: 植物体内引入肥胖基因增产 50%

2021 年 7 月 22 日，北京大学贾桂芳课题组、美国芝加哥大学何川课题组、贵州大学宋宝安课题组合作在 Nature Biotechnology 杂志发表了论文 RNA demethylation increases the yield and biomass of rice and potato plants in field trials。

研究发现在水稻和马铃薯中引入 FTO 基因（动物中著名的肥胖基因），可实现针对 RNA 修饰 m6A 去甲基化，使过表达 FTO 基因的水稻和马铃薯产量和生物量增加了约 50%。

该研究报道了一种表观遗传编辑育种新技术，开辟了一个全新的植物育种方向！

3. Science: 结构生物学揭示 AMPK 活性调节循环机制

AMPK（AMP-activated protein kinase）是细胞内最重要的能量感受器和调节器，其功能失常会导致人体罹患肥胖、糖尿病等疾病。

2021 年 7 月 23 日，美国温安洛研究所（Van Andel Institute）KarstenMelcher 课题组联合中国科学院上海药物研究所徐华强等多个课题组在 Science 杂志发表论文 Structure of an AMPK complex in its inactive, ATP-bound state。

利用 Cryo-EM 技术（冷冻电镜技术），打破蛋白纯化等技术瓶颈，首次解析了 ATP 结合状态下的抑制型 AMPK 复合物的高分辨率结构，从结构生物学角度阐述了 AMPK 的活性调节机制，提出了 AMPK 激活、失活、再激活的循环通路模型！

4. STM：间质细胞在肠道免疫中的作用

炎症性肠病增加了结直肠肿瘤的患病风险。

2021 年 7 月 21 日，上海交通大学 Bio-X 研究院李保界教授等团队在 Science Translational Medicine 杂志上发表研究论文 A resident stromal cell population actively restrains innate immune response in the propagation phase of colitis pathogenesis in mice。

该研究阐明了机体患肠道炎症的负性调节机制，发现间质细胞能够作为临床治疗炎症性肠病的新靶点。

5. Nature: 发现亮氨酸感应器调节规律

mTOR 复合物 1 能够调节细胞生长节律，进而响应氨基酸水平。

2021 年 7 月 21 日，北京大学刘颖及陈杰教授团队在 Nature 杂志上发表研究论文 SAR1B senses leucine levels to regulate mTORC1 signalling，该研究发现 SAR1B 是一种保守的亮氨酸感应器，能够调节 mTOR 复合物 1 从而响应细胞内亮氨酸的水平。

6. Nature：肠道炎症影响嗅觉

在以往的认知中，嗅觉与大脑、鼻子息息相关。

2021 年 7 月 21 日，美国加州诺瓦托巴克老年研究所 Heinrich Jasper 教授团队在 Nature 杂志上发表研究论文 Gut cytokines modulate olfaction through metabolic reprogramming of glia。

该研究表明机体发生肠道炎症时，能够影响中枢神经系统，使得嗅觉变得不再灵敏。同时发现急性肠道感染、衰老过程中积累的慢性肠道炎症均是诱因。

7. Nature：阐明染色体分离的作用机制

精准及时的染色体分离对于维持基因组稳定性意义重大。

2021 年 7 月 21 日，瑞士日内瓦大学 Andreas Boland 教授团队在 Nature 杂志上发表研究论文 Structural basis of human separase regulation by securin and CDK1–cyclin B1。

该研究阐明了 CDK1-cyclin B1 调控人源 separase 活性的分子机制，同时揭示了 separase 抑制 CCC 激酶活性的作用机理。

8. Cell Reports: 研发出新型 RSV 黏膜疫苗

呼吸道合胞病毒 (RSV) 每年造成超过 300 万人住院，目前仍无特异性治疗药物和预防疫苗。

2021 年 7 月 20 日，复旦大学上海公共卫生临床中心鄢慧民和姜世勃教授团队在 Cell Reports 杂志上发表研究论文 A safe and effective mucosal RSV vaccine in mice consisting of RSV phosphoprotein and flagellin variant。

该团队研发出新型 RSV 黏膜疫苗 P-KFD1，抗原为一种重组融合蛋白。P-KFD1 鼻内免疫小鼠，能够显著地降低 RSV 在上呼吸道和下呼吸道的复制。

9.JAMA Internal Medicine：颠覆认知，咖啡能够降低心律失调

全球每天消费约 30 亿杯咖啡，以往报道认为咖啡导致心跳加快。

2021 年 7 月 19 日，加州大学 Gregory M. Marcus 教授团队在 JAMA Internal Medicine 杂志上发表研究论文 Coffee Consumption and Incident Tachyarrhythmias Reported Behavior, Mendelian Randomization, and Their Interactions。

该研究对超过 38 万人进行大规模调查，结果表明一个人每天每多喝一杯咖啡，患心律失常的风险平均会降低约 3%。对于部分人群，咖啡或含咖啡因产品不会扰乱心跳，反而有助于降低心律失常风险。

10.Cell：开发基因疗法阻止视力衰退

当视网膜发生病理变化或受损，视网膜神经节细胞逐渐退化。

2021 年 7 月 22 日，西奈山伊坎医学院 Bo Chen 教授团队在 Cell 上发表研究论文 Preservation of vision after CaMKII-mediated protection of retinal ganglion cells。

该研究开发出一种基因疗法，通过在视网膜神经节细胞中重新激活 CaMKII 关键酶，能对受损的视网膜神经节细胞产生保护作用，为治疗青光眼、视网膜病变等常见致盲疾病提供了新途径。
