产品描述
Anti-V5 Affinity Gel（Anit-V5亲和纯化凝胶）由高质量的鼠源 IgG2b单克隆抗体与Sepharose 4B gel通过供价偶联制备，本产品具有较高的V5标签融合蛋白加载容量（至少为0.5 mg protein/mL凝胶），杂蛋白非特异结合少，可用于带有V5标签的融合蛋白免疫沉淀（IP）和纯化。
推荐使用量：每500 μL蛋白粗体液需使用5 μL凝胶。

运输和保存方法
冰袋运输。未开封的产品在-20℃下可稳定保存1年。禁止在不含甘油的溶液中冻存。

使用方法
1）轻微重悬Anti-V5 Affinity Gel，形成均一溶液。转移10 μL混合液（5 μL gel）至新的离心管中。
2）加入600 μL TBS，轻轻的重悬Anti-V5 Affinity Gel，10,000 rpm离心30 s, 弃上清，重复上述步骤 3-4 次。
注意：尽可能去除上清，但不要吸到凝胶。重复该步骤3-4次。
3）在上述沉淀中加入200-1000 μL细胞裂解液。4℃，轻摇孵育2小时。
4）10,000 rpm离心30 s，上清液转移到新的离心管中保存以后检测。
5）凝胶沉淀用0.5 mL TBS洗3-4 次，直至上清OD280 < 0.05。
6）洗脱：具体选择哪种洗脱条件取决于目的蛋白的特性或下游应用。
A．加入100 μL V5多肽（150 μg/mL）（Cat 20575ES）洗脱V5融合蛋白，4℃轻摇孵育30 min，10,000 rpm离心30 s，收集上清。
B．用于SDS-PAGE电泳：加入10 μL 2×蛋白上样缓冲液，煮沸5 min。10,000 rpm 离心30 s，小心转移上清，用于SDS-PAGE检测。上样缓冲液中SDS会破坏抗体，所以亲和纯化凝胶不能重复使用。
