| 背景介绍 | 白细胞介素21(IL-21)对免疫细胞具有强烈的调节作用，包括可破坏病毒感染或肿瘤细胞的自然杀伤细胞(NK)和细胞毒性T细胞。该细胞因子可诱导靶细胞分裂/增殖。IL-21表达于人活化的CD4+T细胞，但在其他大多数组织中并没有表达。此外，IL-21在辅助性T细胞亚群Th2和Th17细胞以及滤泡性T细胞中表达上调。而且IL-21还表达于调控这些细胞功能的NKT细胞中。
IL-21在免疫炎性疾病中具有病理作用。研究表明存在一个正向自分泌环，可放大并稳定IL-21驱使的、T细胞介导的应答。而且，它还具有抗肿瘤活性，对自身免疫进程具有作用。在临床上，IL-21还可作为潜在的抗肿瘤药物。 |
| 检测原理 | 本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人IL-21抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体，经过孵育，样本中存在的IL-21与固相抗体和检测抗体结合。洗涤去除未结合的物质后，加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后，加入显色底物TMB，避光显色。颜色反应的深浅与样本中IL-21的浓度成正比。加入终止液终止反应，在450 nm波长(参考波长570 - 630 nm)测定吸光度值。 |
