      • 小牛胸腺DNA的制备

        分离出DNA小牛胸腺、猪脾、鱼精子和植物种子的胚胎等生物材料,细胞核含量比例大,因而含有丰富的DNA,是提取DNA的好材料。本实验以小牛胸腺或猪脾为材料。小牛胸腺不仅DNA含最高,而且其脱氧核糖核酸酶(DNase)的活性较低,在制备过程中由此酶所引起的DNA降解损失也较小。为了防止DNase的作用,在用于提取的缓冲液中均含有1mmol/L的EDTA(乙二胺四乙酸)螫合剂,以除去保持 DNase活性所必须的Mg++离子。为防止DNA的变性和降解,操作要尽可能在低温下进行。 本实验通过匀浆
