分离出DNA。 小牛胸腺、猪脾、鱼精子和植物种子的胚胎等生物材料,细胞核含量比例大,因而含有丰富的DNA,是提取DNA的好材料。本实验以小牛胸腺或猪脾为材料。小牛胸腺不仅DNA含最高,而且其脱氧核糖核酸酶(DNase)的活性较低,在制备过程中由此酶所引起的DNA降解损失也较小。为了防止DNase的作用,在用于提取的缓冲液中均含有1mmol/L的EDTA(乙二胺四乙酸)螫合剂,以除去保持 DNase活性所必须的Mg++离子。为防止DNA的变性和降解,操作要尽可能在低温下进行。 本实验通过匀浆
问: 实验室购买的是西格玛产的小牛胸腺DNA,说明书上说保存温度是2~8℃,而由于我们的粗心,我们将其储存在零下20℃的低温储藏箱内,而我们将其配成溶液后,接下来又存储在0~4℃的普通冰箱里。我们用这个保存条件下的DNA做精密度较高的电化学实验,结果发现毫无效果,有指导老师告诉我有可能因为第一步零下20度太低,“冻死了”,请各位虫友支支招,到底应该在什么温度下保存比较好,或者告诉我小牛胸腺
机能,以微量存在于血中,以环核苷酸( cAMP)作为第二信使,可视为胸腺激素( thymin)。其中研究最多的是胸腺素( thy-mosin)。胸腺素为怀特( White)和戈尔茨坦( Goldstein)从小牛胸腺中提取出来的、分子量为 12000道尔顿的蛋白质。能使免疫缺陷病人的 T细胞机能得到恢复,可诱导无胸腺及去胸腺小鼠的 T细胞机能,并可增加小鼠胸腺细胞中的环鸟苷酸。此外,胸腺激素Ⅰ,也是从小牛胸腺中提取出来的多肽,以后进一步提纯成胸腺激素Ⅱ,亦有诱导 T细胞的机能。此激素存在于胸腺
