General description
野生型莓实假单胞菌分离一种金属蛋白酶，其在小亲水性氨基酸处裂解肽键 N-末端。该菌株的一个突变体产生胞内蛋白酶 Asp-N 测序级，分离得到高纯度和特异性蛋白酶。
Specificity
胞内蛋白酶 Asp-N 测序级是一种金属蛋白酶。在 pH 6.0-8.5 的磷酸盐、醋酸盐或 Tris 缓冲液中，特异性裂解天冬氨酸和半胱氨酸的 N 端肽键。如果半胱氨酸还原或烷基化，仅--Asp-X 裂解。
以胰高血糖素为底物，验证了胞内蛋白酶 Asp-N 测序级的特异性。孵育高浓度的胞内蛋白酶 Asp-N 测序级（1 份（按重量）酶和 10 份（按重量）胰高血糖素），以检测痕量杂质，如污染蛋白酶。
热灭活： 煮沸 5 min 使蛋白酶停止。
Application
使用胞内蛋白酶 Asp-N 测序级进行蛋白结构分析和序列分析。
Preparation Note
工作浓度： 2 μg/50μl 双蒸水
1 瓶（= 2μg 酶）溶解于 50μl 双距离。水的缓冲液浓度为 10 mM Tris-HCl pH 7.5。
工作液： 推荐溶剂为双蒸水。
储存条件（工作液）：-15 至-25°C
双联稀释法溶液。水在 2 ~ 8°C 下最多可稳定 1 周，在-15~-25°C 下可稳定约 1 个月；应避免反复解冻。
Reconstitution
用 50μl 双蒸水复溶测序级冻干胞内蛋白酶 Asp-N。得到的缓冲液浓度为 10 mM Tris-HCl，pH 7.5。为避免自溶，孵育温度不得超过 37°C。
