步骤：1. 分离胶和积层胶，制胶板；
注意：灌分离胶时不要加太多。
2. 蛋白变性：准备蛋白样本，Phospho-Raptor (Ser792) Antibody加入等体积的2×上样Buffer稀释，置于沸水中煮10min（预染marker煮5min）
3. 加样：每孔加入20～40μl样品
4. 电泳：置于电泳槽中电泳45min，恒定电流90mA(2快板），如为一块板则电流为50 mA。
5. 取出胶板，用切割刀修好胶；
6. 将胶置于转膜液中浸泡；
7. 按顺序放好下列物质：黑面→海绵→滤纸→胶→NC膜→滤纸（用吸管赶去气泡）→海绵；
8. 置于转膜槽中于，黑面对黑面，加上冰块，加入转膜液；
9. 装好电极，于恒定电压100V下，90min；
10. 丽春红染膜；
11. Phospho-Raptor (Ser792) Antibody用TBST洗去丽春红；
12. 封闭：加入5％脱脂奶粉＋TBST，常温摇床摇1h；
13. 回收封闭液，加入一抗【一抗用5％BSA＋TBS稀释】；
14. 置于4℃摇床摇过夜；
15. 回收一抗，TBST洗膜，10min，共3次；
16. 加入二抗【二抗用5％脱脂奶粉＋TBS稀释】，常温摇床摇1h；
17. 回收二抗，TBST洗膜，10min，共3次；
18. 将膜置于发光液中浸泡约1min；
19. 将膜铺于曝光盒中，于暗室中曝光，洗胶片。
大规模单克隆抗体的生产目前主要有两种方式，Phospho-Raptor (Ser792) Antibody一是腹水制备法，另一种是转瓶培养法。腹水制备法：将杂交瘤细胞株进行体外培养至一定浓度，再将杂交瘤细胞注入小鼠腹腔中，产生腹水，从腹水中纯化单克隆抗体。每只小鼠可以产生3~8 mL腹水，抗体含量可以达到1~10 mg/mL。腹水制备的周期为3~4周。腹水制备法具备成本低、产量高，时间快等优点，是目前主要的大规模抗体生产方式。转瓶培养法：将杂交瘤细胞进行大规模体外培养，从培养基中纯化抗体。抗体含量可以达到20~80 mg/L。转瓶制备法具有抗体品质高、不含小鼠杂蛋白，不含动物病毒等优点。
