Monoclonal antibody against synaptic protein in neuronal cell bodies, dendrites and synapse-rich areas Clone SMI-82 Form Ascites Fluid (contains 0.01M sodium azide) Host Mouse Species Reactivity Mammalian IsoType Synaptic Protein (SMI 82) Monoclonal Antibody IgM Specificity SMI 82 reacts with unidentified synaptic and neuronal components. On Western blots of developing rat brain the antibody reacts with several bands, the most prominent ones at 114,000 and 39,000. On blots of adult rat brain the antibody reacts with a band with molecular weight of 22,000. Blots of adult human brain homogenates indicate reaction similar to the developing rat pattern. SMI 82 visualizes synapse-rich areas of brain and retina in a wide variety of mammalian species. Reaction in adult brain tissue sections from all species examined is primarily with neuronal cell bodies, dendrites and synaptic areas. Axonal staining is faint or absent. In cell cultures, SMI 82 reacts with neuronal cell bodies and dendrites. This IgM antibody reacts with goat antimouse IgG and mouse ClonoPAP¢ç via the light chains common to IgG and IgM. Uses This antibody is effective in immunoblotting, immunocytochemistry and ELISA. Suggested Working Dilution The optimal working dilution should be determined for each specific assay condition. This antibody is sold for laboratory research use only, not for human or in-vivo use. The extent of permissible dilution of SMI 82 beyond those recommended for general application depends upon nature and concentration of the antigen examined, species of the antigen, method of fixation and kind of section examined.

Tissue Preparation: SMI 82 reacts on paraffin, vibratome or frozen tissue sections and with neurons and dendrites in cell cultures. Tissues can be fixed in a variety of paraformaldehyde or formaldehyde containing fixatives. Post-fixation in cold methanol or methanol/hydrogen peroxide facilitates access of SMI 82 to neurons in thick sections of tissue perfused with 4% paraformaldehyde or in tissue cultures.Storage Store at -20°C. Upon initial thawing, apportion into working aliquots and store at -20°C. Avoid repeated freeze-thaw cycles to prevent denaturing the antibody.
