Description 
Monoclonal antibody against myelin basic protein Clone SMI-94 Form Ascites Fluid Host Mouse Species Reactivity Myelin Basic Protein (SMI 94) Monoclonal Antibody Mammalian IsoType IgG1 Specificity SMI 94 reacts with the MBP peptic fragment 70-89 of human myelin basic protein. The antibody reacts with rat, mouse, rabbit, dog, monkey and human MBP. Other species have not been tried, but reactivity may occur over a wide range of mammalian species. SMI 94 detects developing and adult myelin, developing oligodendrocytes and distinguishes oligodendrocytes from astrocytes, microglia, neurons and other cells in brain sections. The combination of SMI 94 with SMI 91 and/or 99 is useful for immunocytochemical studies on the progression of normal and pathologic myelination. Uses This antibody is effective in immunoblotting, immunocytochemistry and ELISA. Suggested Working Dilution The optimal working dilution should be determined for each specific assay condition. This antibody is sold for laboratory research use only, not for human or in-vivo use. Myelin Basic Protein (SMI 94) Monoclonal Antibody The extent of permissible dilution of SMI 94 beyond those recommended for general application depends upon nature and concentration of the antigen examined, species of the antigen, method of fixation and kind of section examined.
Tissue Preparation: SMI 94 reacts on Western blots, paraffin, vibratome and frozen tissue sections and cell cultures. Tissues can be fixed in a variety of paraformaldehyde or formaldehyde-containing fixatives. To insure penetration of antibody into myelin it is necessary to pretreat frozen or vibratome sections with cold methanol (30 min) or detergent. Reaction on formalin-fixed, paraffin-embedded sections may be enhanced by autoclaving deparaffinized sections in distilled water for 10 min (Shin et al, Lab Invest 64:693,1991) or by boiling sections or tissue blocks immersed in tris buffered saline, pH 9.0, in a microwave oven for 15 min (Evers, Uylings, J Neurosci Meth 72:197, 1997).Storage Store at -20°C. Upon initial thawing, apportion into working aliquots and store at -20°C. Avoid repeated freeze-thaw cycles to prevent denaturing the antibody.
