Product Overview
Stemolecule™ IDE-1 was identified in a high throughput screen as an inducer of definitive endoderm (IDE) from embryonic stem (ES) cells1. IDE-1 is able to generate Sox17+/FoxA2+ cells to an equal or greater efficiency than that of protein (i.e. Activin A or Nodal) inducers of definitive endoderm in mouse or human ES cell cultures1. It has an EC50 of 125.5 nM and functions in part via activation of TGF-β signaling, as evident by Smad2 phosphorylation1. Endoderm chemically induced by IDE-1 can continue to differentiate into pancreatic progenitors in vitro and contribute to gut tube formation in vivo when injected into the developing gut tube of mouse embryos1.

Stemolecule™ IDE-1Solubility Soluble in DMSO (100 mM). For a 10 mM concentrated stock solution, reconstitute the compound by adding 653 µl of DMSO to the entire contents of the vial. Note: for most cells, the maximum tolerance to DMSO is <0.5%.
Storage Store at 4°C. Protect from light. Following reconstitution, store aliquots at -20°C.
Stability Stock solutions stable at -20°C for 6 months.
Quality Control The purity of IDE-1 was determined by HPLC analysis. The accurate mass and structure of IDE-1 were determined by mass spectrometry and NMR, respectively. Cellular toxicity of IDE-1 was tested on mouse ES cells.
