The Drosophila HP1 protein, product of the Su(var)2-5 locus, is a dose-dependent regulator of gene silencing. HP1 interacts with compacted chromatin (heterochromatin) via its chromodomain (CD). HP1 antisera staining of polytene chromosomes decorates pericentric heterochromaticn and a banded heterochromatic pattern on the small 4th chromosome. The HP1 protein is also known to interact with a number of transcriptional regulators via its chromoshadow domain (CSD), suggesting that HP1 is a bridging factor that couples particular chromatin structures with effects on gene expression. This antibody was raised the peptide sequence, YAVEKIIDRRVRKGKVEYYLKWKG, corresponding to amino acids 24-47 of the Drosophila HP1 protein within the chromodomain (CD) of HP1. It has been shown to specifically stain Drosophila polytene chromosomes.
Uses
This antibody is effective in immunostaining.
Suggested Working Dilution
Drosophila HP1 Polyclonal Antibody The optimal working dilution should be determined for each specific assay condition. This antibody is sold for laboratory research use only, not for human or in-vivo use. Covance antibodies may not be resold or modified for resale without prior written approval.
Immunostaining: 1:200
Storage
Store at -20°C. Upon initial thawing, apportion into working aliquots and store at -20°C. Avoid repeated freeze-thaw cycles to prevent denaturing the antibody. For long-term storage, keep the antibody at -80°C.
