㊣Annexin Ⅴ凋亡检测试剂盒
在正常细胞中,磷脂酰丝氨酸只分布在细胞膜脂质双层的内侧,细胞发育凋亡早期,膜磷脂酰丝氨酸(PS)由脂膜内侧翻向外侧。Annexin Ⅴ是一种磷脂结合蛋白,与磷脂酰丝氨酸有高度亲和力,故可通过细胞外侧暴露的磷脂酰丝氨酸与凋亡早期细胞的胞膜结合。因此Annexin Ⅴ被作为检测细胞早期凋亡的灵敏指标之一。在细胞发育凋亡时,膜磷脂酰丝氨酸外翻的发生早于细胞核的变化。因为细胞坏死时也会发生膜磷脂酰丝氨酸外翻,所以Annexin Ⅴ常与鉴定细胞死活的核酸染料(如PI)合并使用,来区分凋亡细胞与死亡细胞。

以Annexin Ⅴ FITC试剂盒为例进行以下说明:
试剂盒组份:
● FITC标记的Annexin Ⅴ
● PI染料(20ug/ml)
● 结合缓冲液
注意事项:
● 保证细胞的状态,避免出现过多的死亡细胞
● 操作过程避免过度用力,以免出现机械性细胞死亡
● 使用去离子水准备所需溶液
● 2-4℃避光保存试剂盒
● 使用一次性吸头,避免出现交叉污染
Annexin V Unlabeled Recombinant Protein基本操作
● 按照既定程序诱导细胞凋亡
● 按照说明书配制所需容溶液
● 讲细胞悬浮在1×结合缓冲液中,浓度调整为2-5×105/ml
● 将细胞悬浮液和FITC标记Annexin Ⅴ混合,温室孵育10分钟
● 离心,PBS洗涤细胞;使用1×结合缓冲液重悬细胞
● 使用PI染色细胞(终浓度为1ug/ml)
● 使用流式细胞仪分析
