TGM2 / TGC / Transglutaminase-2抗体背景综述
Protein-glutamine gamma-glutamyltransferase 2, also known as Tissue transglutaminase, Transglutaminase C, Transglutaminase-2, and TGM2, is a member of the transglutaminase superfamily. Human TGM2 is implicated in the pathogenesis of several neurodegenerative disorders including Alzheimer's, Parkinson's and expanded polyglutamine (polyQ) diseases. TGM2 promotes formation of soluble and insoluble high molecular weight aggregates by catalyzing a covalent linkage between peptide-bound Q residues in polyQ proteins and a peptide-bound Lys residue. TGM2 plays a role in cell growth and survival through the anti-apoptosis signaling pathway. Its expression is involved with tumor growth. The expression of TGM2 plays an important role in doxorubicin/cisplatin resistance in breast and ovarian cancer. Down-regulation and over-expression of TGM2 in non-small cell lung cancer (NSCLC) cell suggested a positive correlation of cisplatin sensitivity and TGM2 inhibition. Good responders of cisplatin in NSCLC could be identified by the promoter methylation of TGM2. TGM2 is also useful as a predictive marker for patient prognosis and may be a novel therapeutic target for colorectal cancer (CRC).
