Produced in rabbits immunized with purified, recombinant Human Complexin-2 / CPLX2 (rh Complexin-2 / CPLX2; Catalog#11927-H07E; Q6PUV4-1; Asp 2-Lys 134). Complexin-2 / CPLX2 specific IgG was purified by Human Complexin-2 / CPLX2 affinity chromatography.
Complexin-2 / CPLX2抗体 Background
Complexin-2 (CPLX2) belongs to the complexin / synaphin family, is a soluble pre-synaptic protein believed to regulate neurotransmitter release from pre-synaptic terminals. Complexins regulate the activity of soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE) complexes necessary for vesicle fusion. Complexin 1 (CX1) and complexin 2 (CX2) are presynaptic proteins that modulate neuro-transmitter release and are used as markers of inhibitory and excitatory synapses, respectively. CPLX2 is localized in pre-synaptic terminals in mature brain. Immunofluorescence microscopy revealed complexin-2 localized along the apical plasma membrane consistent with a role in secretion. Accordingly, complexin-2 was found to interact with vesicle-associated membrane protein (VAMP) 2, syntaxins 3 and 4, but not with VAMP 8 or syntaxin 2. Introduction of recombinant complexin-2 into permeabilized acini inhibited Ca (2+)-stimulated secretion in a concentration-dependent manner with a maximal inhibition of nearly 50%. CPLX2 participates in the Ca (2+)-sensitive regulatory pathway for zymogen granule exocytosis. The G71-P89 region of CPLX2 is essential and sufficient for preferential axonal distribution. Previous studies have reported that the protein and mRNA levels of the complexin-2/CPLX2 was decreased in patients with schizophrenia. Complexin-2 is involved in synaptogenesis and the modulation of neurotransmitter release. Additionally, it is a key player in normal neurological function, and that downregulation of complexin-2 could lead to changes in neurotransmitter release sufficient to cause significant behavioural abnormalities such as depression.
