| 描述 | Linaclotide developed for the treatment of constipation-predominant irritable bowel syndrome (IBS-C) and chronic constipation, and is a potent and selective guanylate cyclase C agonist. |
| 体外活性 | Linaclotide 通过作用于肠腔膜上的鸟苷酸环化酶-C受体，增加氯离子和碳酸氢盐向肠道的分泌，并抑制钠离子的吸收，从而增加水分向肠腔的分泌并改善排便；该化合物在体循环中的吸收极少[2]。体外实验中，Linaclotide 能浓度依赖性地抑制[125I]-STa对来自野生型小鼠肠粘膜膜的结合。相比之下，[125I]-STa与来自GC-C基因敲除小鼠的这些膜的结合显著降低。在体外空肠液中孵育30分钟后，Linaclotide 完全降解[1]。 |
| 体内活性 | Linaclotide 在慢性便秘患者中显著增加每周自发性排便次数及完全自发性排便次数（CSBMs），同时减轻腹痛[2]。药代动力学分析显示其口服生物利用度极低（0.10%）。在肠分泌和传输模型中，linaclotide 在野生型小鼠中展现出显著的药理作用，但在GC-C null小鼠中则不然：通过外科手术结扎的空肠环路内增加液体分泌，伴随着循环鸟苷酸-3',5'-单磷酸（cGMP）水平的升高和胃肠传动的加速[1]。 |
