产品描述
肿瘤坏死因子α(TNF alpha)是一种主要由巨噬细胞和单核细胞产生的促炎细胞因子，能够直接杀伤肿瘤细胞而对正常细胞无明显毒性。TNF alpha在细胞内组装，形成一个非共价连接的同源三聚体，并在细胞表面表达。TNF alpha参与正常炎症反应和免疫反应，可以通过协同调节其它细胞因子的产生、细胞存活和死亡来协调组织的稳态。
本试剂盒采用夹心法检测TNF alpha含量，试剂盒中有TNF alpha Standard以及两株特异性识别TNF alpha的单克隆抗体，其中一株抗体偶联Eu(供体，Anti-TNF alpha-Eu)，另一株抗体偶联A3(受体，Anti-TNF alpha-A3)。当两株抗体同时与TNF alpha结合时，Anti-TNF alpha-Eu和Anti-TNF alpha-A3距离靠近，可发生荧光共振能量转移(FRET)。使用340 nm的激发光激发荧光供体，供体发射620 nm的光，此620 nm的光激发荧光受体，受体发射出665 nm的光。样本中待测TNF alpha浓度与FRET信号值(665 nm/620 nm的光强度比值)成正比。
