实验方案
此通用协议适用于 ABI7500 FAST qPCR 仪器。
准备 NED 校准板:
1.将 1 μL 的 10,000X NED 染料 qPCR 校准溶液与 10 mL 的无核酸酶水或 qPCR 缓冲液混合,制备 NED 染料 qPCR 校准工作溶液。请注意,染料浓度必须根据仪器的校准要求进行优化。
2.在反应板的所有孔中加入 20 μL 的 NED Dye qPCR 校准工作溶液。
3.用光学胶盖密封板
4.以 < 1500 rpm 的速度将板离心 2 分钟
5.检查板的每个孔中的液体是否位于孔的底部。如果没有,请再次以稍高的 rpm 和更长的时间将板离心
