文献和实验获细胞，加入适量细胞裂解缓冲液（含蛋白酶抑制剂），冰上裂解 30 min，细胞裂解液于 4 ℃，最大转速离心 30 min 后取上清； 取少量裂解液以备 Western blot 分析，剩余裂解液加 1 μg 相应的抗体加入到细胞裂解液，4 ℃ 缓慢摇晃孵育过夜； 取 10 μl protein A 琼脂糖珠，用适量裂解缓冲液洗 3 次，每次 3 000 rpm，离心 3 min； 将预处理过的 10 μl protein A 琼脂糖珠加入到和抗体孵育过夜的细胞裂解液中 4 ℃ 缓慢摇晃孵育
样本准备 蛋白提取 所需仪器：匀浆器、RIPA 裂解液、PMSF（蛋白酶抑制剂）、磷酸酶抑制剂、剪刀、镊子，消毒酒精棉、PBS、移液器、离心机。 裂解液配制——RIPA 裂解液：PMSF=100：1。 1） 组织处理：首先用酒精棉消毒剪刀和镊子，取适量的组织于匀浆器中，用剪刀剪碎加适量的裂解液冰水中匀浆 1-4 min。匀浆结束后吸取溶液于 EP 管中保存备用。（含血液较多的组织可先用 PBS 清洗后再匀浆） 2） 细胞处理： A、 贴壁细胞处理：去掉培养基用 PBS 清洗后加入适量的裂解
