| 产品描述： | The protein encoded by this gene belongs to the highly conserved cyclin family, whose members are characterized by a dramatic periodicity in protein abundance through the cell cycle. Cyclins function as regulators of CDK kinases. Different cyclins exhibit distinct expression and degradation patterns which contribute to the temporal coordination of each mitotic event. This cyclin forms a complex with and functions as a regulatory subunit of CDK2, whose activity is required for cell cycle G1/S transition. This protein accumulates at the G1-S phase boundary and is degraded as cells progress through S phase. Overexpression of this gene has been observed in many tumors, which results in chromosome instability, and thus may contribute to tumorigenesis. This protein was found to associate with, and be involved in, the phosphorylation of NPAT protein (nuclear protein mapped to the ATM locus), which participates in cell-cycle regulated histone gene expression and plays a critical role in promoting cell-cycle progression in the absence of pRB. |

文献和实验microRNA-195通过直接影响靶基因Cyclin D1和Cyclin E1的表达 抑制了人神经胶质瘤细胞的增殖
膜母细胞瘤（pRb）和增殖细胞核抗原（PCNA）的形成。相反，抑制了miR-195的作用后，神经胶质瘤细胞的增殖速度加快，S期细胞的比例升高，G1/G0期的细胞数减少，提高细胞的非依赖型锚定生长能力，促进细胞中pRb和PCNA的磷酸化。此外，我们还发现miR-195通过直接作用于Cyclin D1和Cyclin E1的mRNA 3’UTR区下调Cyclin D1’和Cyclin E1的表达，抑制了神经胶质瘤细胞的增殖。综上所述，该实验结果表明miR-195在抑制神经胶质瘤细胞增殖的过程中发挥了重要的作用，并阐明

的抑癌作用。他克隆了 Rb 全长的 cDNA ，发现其产物是一种长 928 氨基酸，分子量为 105Kda 的蛋白质。 Rb 的分子功能是什么呢？它主要是可以和其它各种蛋白相互作用，其中包括肿瘤抗原： SV40 T 抗原，腺病毒 E1A 抗原，人类乳头瘤病毒 E7 抗原，可结合到 RB 的 379~792 残基区域。一种可能是部分致癌蛋白由于和 RB 产物的相互作用其致癌的作用受到了抑制。另外 RB 还有抑制细胞增殖的作用。
