Description :CD51/CD61 is an integrin complex known as α β . It is expressed at high levels on osteoclasts,endothelial cells, and melanoma cells and at low levels on platelets and macrophages. CD51 isa heterodimer composed of disulfide-linked 125 kD and 24 kD proteins. CD61 is also amember of the integrin family known as gpIIIa or β integrin. It is a 110 kD common β subunit ofCD51/CD61 or CD41/CD61 complex. CD51/CD61, also known as the vitronectin receptor,mediates the binding of platelets to immobilized vitronectin without prior activation. Otherligands include RGD-containing proteins such as fibrinogen, fibronectin, von Willebrand factor(vWf), laminin, thrombospondin and the neural adhesion molecule L1. CD51/CD61 alsomediates cell-cell adhesion via interaction with CD31. CD51/CD61 acts as an activationindependent receptor for platelet attachment and spreading on vitronectin and other RGDcontaining proteins, including matrix components. The 23C6 antibody has been reported to beuseful for blocking studies.

当FITC在碱性溶液中与抗体蛋白反应时,主要是蛋白质上赖氨酸的r氨基与荧光素的硫碳胺键(thiocarbmide)结合,形成FITC-蛋白质结合物,即荧光抗体或荧光结合物。一个IgG分子中有86个赖氨酸残基,一般最多能结合15~20个,一个IgG分子可结合2~8个分子的FITC,其反应式如下FITC-N=C=S + N-H2-蛋白质 → FITC-NS-C-N-H2-蛋白质常用Marsshall(1958)法标记荧光抗体,也可以根据条件采用Chadwick等标记法或Clark
