Isolation of Yeast DNA

The problems often experienced with DNA isolated from yeast are mostly owing to the impurities remaining in DNA isolated according to certain isolation protocols. Incomplete cutting of chromosomal DNA with restriction endonucleases, or poor transformation of Escherichia coli with plasmid DNA from yeast are the difficulties most frequently encountered. The procedures given herein have been chosen since they have used successfully by many workers to obtain DNA that does not give these problems.
