MassPREP自动酶切仪操作步骤

一 开机前检查：

1 检查仪器台面（DECK）上所有的实验材料（Labware）。

2 检查SystemWater 水桶的水位。

3 检查恒温循环水浴（Chiller）水箱的水位，并定期更换或填充Chiller 中的循环水。

4 倒掉废液桶中的废液。

二 开机步骤：

打开Chiller、Heater、仪器及计算机电源，进入WinPREP程序 ：

1 仪器初始化

点击菜单：[Utilites]→[Setup]→[Instrument]→[Initialize]仪器进行初始化。

2 系统管路冲洗、除气泡

点击菜单：[Utilites]→[Diagnostics Test]→[Flush and Wash Tips]；

放置Labware(washbowl+1Through)在仪器平台的G13位置，排液管接至废液桶，按提示执行程序：

Deck layout OK,

run test now。 √

Flush Wash Volumes

Enter the Flush Volume(vl): 3000

Enter the Wash Volume(vl) : 3000

>start,仪器开始冲洗管路中的气泡，要多次冲洗直至管中完全没有气泡。

3 执行自动酶切程序

点击菜单：[File]→[Open]；

打开所要执行的自动酶切操作文件：

按文件图示位置放置所需的Labware。（注意：盖子应放在96 well ABgene PCR 上； 所有实验器皿应放入准确位置）， 按要求配置试剂，装入指定试剂盒（瓶）；

点击工具栏：[Execute Test]。弹出[Test Startup Page]窗口, →点击[Reset Tip Boxes],（Lid on PCR(1-96) ,Tips=1），“Fill”为添满，“Empty”为清空，

>Start，开始执行程序。

注意事项：

1． 为避免角蛋白和其它污染，在移动labware 、dispensable tips、试剂或样品时，请戴手套操作，在仪器运行过程中非仪器操作人员请远离仪器。

2． 整个操作过程中的水为除去气泡的去离子水。

3． 检查冷却循环仪是否打开和是否达到安全水位；检查系统液体的水平面是否达到标志线（1/2桶的体积）；检查废水桶的液面，及时排水；检查各种实际是否准确配置及正确放入指定位置。

4． 执行程序前，一定要对照所选执行程序检查所有Labware的位置是否正确

5． 盖子（lid）应放在96 well ABgene PCR （1-96）上。

6． 运行FlushLiq ，尽可能的去除系统中可能存在的气泡。

7． 在[Reset Tip Boxes]时，要确定盖子（lid）位置和枪头（Tips）数量与实际位置和数量相符

8． 在仪器运行过程中如有以外情况发生，请速按仪器正前方紧急停止按扭（红色）。

9． 在仪器关机不用时，请用仪器罩遮严以防止灰尘。

10． 在每次操作完成后请自觉登记，并保证操作台面的整洁。
