The Human cluster of differentiation 63 (CD63) ELISA Kit is an enzyme-linked immunosorbent assay for the quantitative detection of human CD63 in many biological fluids, including serum, plasma, and tissue homogenates. This assay employs the Sandwich-ELISA technique, in which CD63 in the samples or standards is sandwiched between pre-coated an anti-human CD63 and Biotin-labeled CD63 antibody. The Ag-Ab-Ag immune complex is labeled with HRP-avidin and then develops a color reaction after the addition of the TMB substrate. The intensity of the color is positively correlated to the amount of CD63 in the sample. 一、试剂

2.试剂的准备：先将试剂盒先从冰箱中拿出来，在室温下放置20-30 min后，再进行测定，使试剂盒在使用前与室温平衡，这样做的目的能使反应微孔内的温度较快地达到所需的温度，以满足后面的测定需求。
二、样本
1.内源性干扰因素：包括类风湿因子、补体、高浓度的非特异免疫球蛋白、异嗜性抗体、某些自身抗体等；
类风湿因子的控制方法：
①用F(ab)2替代完整的IgG；
②标本用联有热变性IgG的固相吸附剂处理；
③检测抗原时，可加入到标本稀释液中使RF降解；
补体的控制方法：
①用EDTA稀释标本；
②56℃ 30 min加热血清使C1q灭活；
2.外源性干扰因素：包括标本溶血、细菌污染、保存时间过长或凝固不全等；
控制方法：采血时规范操作，采集的血液勿用力震荡，以防溶血；收集标本时采用无菌试管，经检测后再低温冻存；保存时间不宜过久，检测时尽量采用新鲜标本；对于凝固不全的血标本可适当加入抗凝剂，并放入37℃水中1 h，待充分凝固后再离心分离血清。
