DNA沉淀剂：
原则：不与DNA反应，对其无影响。
1.纯乙醇（二倍体积比）；效率高，需要放置的时间短，在公司用Kit提取的时候，没有放置直接进行下一步；确定是价格相对昂贵。
2.95%乙醇，价格相对于纯乙醇便宜，但是会损失DNA；
3.异丙醇（0.6倍体积）；
（1）普通细胞上清液体，静止15-30min即可沉淀；
（2）对于含量低，或者组织（比如我现在经常提取的小鼠的肝脏中的Total和cccDNA），则最好在-20℃放置2h或者以上，
本人提取肝脏组织DNA时候，最长时间放置了三天，对结果并无影响；上周提取细胞中cccDNA的时候，室温放置了15min，结果提出的DNA含量也挺高的，而且之后的Southern结果也很成功。我猜测可能是含量高，所以时间短点也可以，但是如果时间不是很着急，还是按照规定的时间来做比较保险。

作用及原理：
⒈溶液I—溶菌液：
溶菌酶：它是糖苷水解酶，能水解菌体细胞壁的主要化学成分肽聚糖中的β-1,4糖苷键，因而具有溶菌的作用。当溶液中pH小于8时，溶菌酶作用受到抑制。
葡萄糖：增加溶液的粘度，维持渗透压，防止DNA受机械剪切力作用而降解。
EDTA：（1）螯合Mg2＋、Ca2＋等金属离子，抑制脱氧核糖核酸酶对DNA的降解作用（DNase作用时需要一定的金属离子作辅基）;（2）EDTA的存在，有利于溶菌酶的作用，因为溶菌酶的反应要求有较低的离子强度的环境。
⒉溶液II－NaOH－SDS液：
NaOH：核酸在pH大于5，小于9的溶液中，是稳定的。
但当pH＞12或pH＜3时，就会引起双链之间氢键的解离而变性。
BspLI (NlaIV) (10 U/µL)在溶液II中的NaOH浓度为0.2mo1／L，加抽提液时，该系统的pH就高达12.6，因而促使染色体DNA与质粒DNA的变性。
SDS：SDS是离子型表面活性剂。它主要功能有：
（1）溶解细胞膜上的脂质与蛋白，因而溶解膜蛋白而破坏细胞膜。
（2）解聚细胞中的核蛋白。
（3）SDS能与蛋白质结合成为R-O-SO3-…R＋-蛋白质的复合物，使蛋白质变性而沉淀下来。但是SDS能抑制核糖核酸酶的作用，所以在以后的提取过程中，必须把它去除干净，防止在下一步操作中（用RNase去除RNA时）受到干扰。
根据不同浓度的氯化钠溶液对DNA和蛋白质的溶解度不同原理，可以在提取不同类型DNA使用中加以利用。
1.Nacl是常用的盐析材料之一，盐析即在高浓度Nacl溶液中，蛋白质析出的现象。原理如下
（1）Nacl溶液与蛋白质争夺水分子，破坏蛋白质胶体颗粒表面的水膜；
（2）Nacl溶液大量中和蛋白质颗粒上的电荷，从而使水中蛋白质颗粒积聚而沉淀析出；
2.不同浓度下DNA分子构象（三维结构）不同，不同的构象溶解度也不同。
（1）当NaCl溶液的浓度为0.14mol/L时，DNA溶解度最小，因此改变NaCl溶液的浓度既能使DNA溶解也能使其析出；
（2）DNA在不同浓度的氯化钠溶液中随着氯化钠的浓度不断升高；
3.所以可以使用适当的高浓度Nacl溶液，使得蛋白质析出，而DNA溶解度较大，通过高速离心，取上清并去除沉淀，进而去除蛋白质。
