请勿将本品置于-20 ℃或者液氮中保存。 质量控制 除农杆碱型 Ri 质粒外，无外源质粒 DNA 残留； 电转法转化效率≥105 cfu/μg DNA 使用方法 1. 将新的 2 mm 电击杯插入冰中预冷；或者将储存于 75%乙醇中的 2 mm 电击杯和杯盖取出，在超净工 作台中将其倒置于干净的吸水纸上，使乙醇充分挥发，然后插入冰中预冷； 2. 取-80℃保存的农杆菌感受态插入冰中待其融化，加入 0.01-1μg 质粒 DNA（体积不大于 5μl），用 200μl 枪头将感受态-质粒混合物快速转移到预冷的电击杯中，盖上杯盖； 3. 开启电转仪，设置参数（2500 V ，200 Ω，25μF），将电击杯快速放入电转槽中，电击完成后快速插 入冰中，加入 900μl 无抗生素的 YEB 并将感受态细胞转移到无菌空管中，28 ℃振荡培养 2~3 小时。 （注：此电转化参数为 Bio-rad 推荐，使用者也可使用电转仪所推荐的 Protocol 进行操作） 4. 6000 rpm 离心一分钟收菌，留取 100μl 左右上清将菌体轻轻吹匀，并涂布于含相应抗生素的 YEB 平板 上，倒置放于 28℃培养箱培养 2~3 天。 注意事项 1. 加入质粒时体积不应大于感受态体积的 1/10； 2. 电转感受态细胞中不可混入气泡，转化高浓度的质粒可相应减少最终用于涂板的菌量； 3. 平板上阳性克隆密度不宜过大，为了获得大的菌落，应减少质粒用量或最终用于涂板的菌量； 4. Ar.Qual 具有链霉素、氯霉素抗性，不可用于具有链霉素抗性或氯霉素抗性质粒的转化。
