Description: Affinity purified rabbit polyclonal anti-acetylated lysine antibodies. Developed using a unique technique that utilize the acetylated KLH as immunogen and the acetylated lysine-specific antibodies were specifically purified with immobilized acetylated lysine on agarose. The purified antibody was conjugated to d-Biotin via amine acylation. Direct label of primary antibody will avoid the use of secondary antibodies therefore eliminating the interference of the 2nd antibody-conjugates.
Species: Rabbit
Formulation: 250 μg/mL antibody stored in PBS, pH 7, with 50% glycerol.
Storage & Stability: Store product at –20 ℃. Three years from date of shipping.
Immunogen: Acetylated KLH conjugates.
Purification: The antibody was purified on acetyl-lysine agarose.
Conjugation: Acylation with NHS-Biotin, Biotin/Ab molar ratio: 10:1.
Application(s): The antibody recognizes proteins acetylated on lysine residues.
Tested: acetylated histone, acetylated BSA, and acetylated MBP, no reaction to the non-acetylated proteins.
Scientific Description: DNA transcription can not take place unless DNA is unwound from the nucleosomes. The cell unwinds the DNA by acetylation of lysine residues within the proteins. It has been suggested that acetylation of non-histones proteins (e.g., transcription factors) and histones may be involved. Acetylation of the proteins may result in signal transduction within the chromatic domains.
