超敏ECL化学发光试剂盒是一款超灵敏ECL化学发光底物,可与酶标二抗上偶联的辣根过氧化物酶发生化学反应发出荧光,从而可以通过X光片压片或其它适当荧光成像设备检测。超敏ECL化学发光试剂在高敏 ECL的基础上加入了二段增强体系,荧光信号强度较高敏ECL提高了5-10倍且信号时间无明显缩短,超敏 ECL同样适用于常规Western blot实验,应用于信号弱的指标效果更佳理想,对于抗体剥脱重新杂交的印迹膜依然可以获得很好的重现性。

4℃避光保存,半年有效。如果长期不用,可以-20℃保存。-20℃可以保存更长时间。
注意事项:
1. A液和B液取用时一定要更换吸头,以免相互混入导致试剂失效;
2. A液和B液使用后,请盖紧瓶盖放回4℃冰箱,避免长期暴露导致试剂失效;
3. 工作液现用现配,一旦跟印迹膜反应请在20min内检测荧光信号;
4. A液和B液均对人体有害,操作时请佩戴一次性手套和口罩。

操作流程:
1. 将HRP标记二抗或HRP标记一抗孵育结束后的印迹膜用TBST漂洗10min×4次;
2. 按照A液:B液1:1的比例配制相应体积的底物工作液,每10cm²印迹膜约需要1.25ml底物工作液;
3. 将印迹膜表面的TBST在干净吸水纸上沥干,平铺在一张干净的保鲜膜上;
4. 将事先配好的底物工作液均匀的洒在印迹膜表面,反应2-5min后,将表面底物工作液倾去;
5. 将印迹膜夹在两层保鲜膜之间,用玻璃棒赶出多余液体,或用X光片压片曝光,或放入成像仪内拍照。
