产品名称:饱和油红O染液
产品概述:本产品主要用于脂质组织染色,染色结果是:组织细胞中脂滴呈橘红色,核呈蓝色。
注意事项:
1、冰冻切片需要先固定再染色,如组织已固定可直接晾干后进行染色
2、工作液必须现配现用,根据样本量适当配置,避免浪费。
3、整个染色过程中注意动作不要太大,以免脂肪丢失或移位。
4、染色结果随时间会褪色,封片后应尽快观察及拍照,可短期4℃保存。

操作流程:
1、取出冰冻切片,常温晾干。
2、分化:取出切片,停留3s,先经60%异丙醇分化3s、更换新的60%异丙醇在分化5s,再依次浸入2缸纯水中浸洗,每缸各10s。
3、油红O染色:切片轻轻浸入油红O工作液浸染8-10min(加盖避光)。
4、分化:取出切片,停留3s,先经60%异丙醇分化3s、更换新的60%异丙醇在分化5s,再依次浸入2缸纯水中浸洗,每缸各10s。
5、苏木素染色:取出切片,停留3s后浸入苏木素复染3-5min,3缸纯水浸洗,各5s、10s、30s,盐酸酒精分化,蒸馏水洗,返蓝液返蓝,蒸馏水冲洗 。
6、封片:甘油明胶封片。
7、显微镜镜检。
【注】配好的油红O工作液要在2个小时内用,不能放置时间过长。
