TNIK (TRAF2 and NCK interacting kinase) is a protein kinase belonging to the germinal center kinase family. It consists of an N-terminal kinase domain. It is localized in the nucleus. TNIK is highly involved in the cytoskeleton assembly and neuronal dendrite extension. In cytoskeleton arrangement, it directly interacts with Rap2 for the proper distribution and spreading of cells. It also stimulates c-Jun N-terminal kinase for the cytoskeleton regulation. TNIK acts as transcription activator during Wnt signaling, which is an essential step for several physiological processes, including cell proliferation, differentiation, motility, apoptosis. It also plays an important role in the intestinal crypts and colorectal cancer cells. It has been suggested that TNIK can be a potential pharmacological kinase inhibitor for the treatment of colorectal cancer.
