关于Annexin A7：
Annexin A7 (ANXA7) is a member of the annexin family of calcium-dependent phospholipid binding proteins. Annexin A7 has a unique, highly hydrophobic N-terminal domain and a conserved C-terminal region. The C-terminal region is composed of alternating hydrophobic and hydrophilic segments. Annexin A7 is a calcium/phospholipid-binding protein with diverse properties including voltage-sensitive calcium channel activity and promotes membrane fusion and is also involved in exocytosis.

  本酶为一种新型的高特异性高活性Fab切割酶，通过大肠杆菌表达，经多步纯化获得。该酶能对人、鼠、绵羊、山羊等物种不同亚型的免疫球蛋白IgGs进行快速单位点酶切。该酶的酶切位点位于免疫球蛋白IgG铰链区的上方，酶切产物为Fab片段和二聚体Fc片段。与木瓜蛋白酶，无花果蛋白酶，胃蛋白酶等相比，FabCutter I Protease具有酶切位点单一，切割快速等优点。
FabCutter酶切体系缓冲液的最适pH值为6.5-8.0（表一），最适反应温度为37℃，另外该酶发挥活性需要还原剂（表二）。

活性定义：在10mM Tris, 150mM NaCl,2mM DTT,pH 7.4的反应体系内，37℃反应1小时，一个单位的酶能将1μg的人源IgG切割95%以上。

应用实例：

One unit cleaves≥95% of 1μg human IgG1 when incubated in 10mM Tris,150mM NaCl,2mM DTT,pH 7.4 at 37℃ for 1 hour as monitored by SDS-PAGE.

溶解方法：
1、溶解前低速离心，使冻干粉末聚集于管底。
2、用无菌ddH2O溶解，1000U酶加入20μl无菌ddH2O，5000U酶加入100μl无菌ddH2O，配制成酶活性为50U/μl的溶液。
3、低速离心，使溶液汇聚于管底。

操作方法：
1、按照溶解说明将冻干粉溶解，配制成为50U/μL的溶液。
2、待酶切的抗体溶解在反应缓冲液里，建议浓度为0.5-10mg/ml。
3、按照1U酶切1μg IgG的量将酶加到反应缓冲液里，加入还原剂激活酶的切割反应，通常在中性pH条件下加入1-5Mm DTT或者TCEP。
4、37℃酶切1个小时，取部分样品跑SDS-PAGE检测切割效果。
注：可使用本品中的Human IgG抗体做为底物测试酶切效果，用70μl ddH2O配制成1mg/ml的蛋白溶解，取50μl加入1μl的酶。酶切前后取样跑SDS-PAGE。

储存条件：-20℃。

Transaldolase (TALDO1) belongs to the transaldolase family of Type 1 subfamily. TALDO1 is expressed selectively in oligodendrocytes of the brain. Retinol-binding proteins (RBP) are a family of proteins with diverse functions. They are carrier proteins that bind retinol. Retinol and retinoic acid play crucial roles in the modulation of gene expression and overall development of an embryo. However, deficit or excess of either one of these substances can cause early embryo mortality or developmental malformations. Regulation of transport and metabolism of retinol necessary for a successful pregnancy is accomplished via RBP. Retinol binding proteins have been identified within the uterus, embryo, and extraembryonic tissue of the bovine, ovine, and porcine, clearly indicating that RBP plays a role in proper retinol exposure to the embryo and successful transport at the maternal-fetal interface.

Vitamin K-Dependent Protein C (PROC) is a serine protease that belongs to the peptidase S1 family. Human PROC is synthesized as a single chain precursor, which is cleaved into a light chain and a heavy chain held together by a disulfide bond. PROC is expressed in plasma and liver. PROC contains one peptidase S1 domain, one Gla (γ-carboxy-glutamate) domain and two EGF-like domains. PROC is a vitamin K-dependent serine protease that regulates blood coagulation by inactivating factors Va and VIIIa in the presence of calcium ions and phospholipids. Defects in PROC are the cause of thrombophilia due to protein C deficiency, autosomal dominant (THPH3) and autosomal recessive (THPH4).
