General description
NBT是一种发色和可溶性物质。它是碱性磷酸酶（AP）的底物，并在其作用下转化为靛蓝/紫色染料。

Physical form
该溶液（100 mg/ml）的溶剂为70%二甲基甲酰胺(DMF) (v/v)。NBT溶液的颜色介于黄色和棕色之间，具体取决于所用批次。
注意：颜色不影响试剂的质量或功能。在某些情况下，可能产生沉淀，在37℃下短暂加热即可使沉淀溶于溶液。
Preparation Note
工作溶液：染色溶液的制备
• 临用前配制新鲜溶液！对于除DIG标记系统以外的所有应用：将50 μl NBT溶液和37,5 μl BCIP加至10 ml 0.1 M Tris-HCl中，pH 9.5 (20 °C)，含0.1 M NaCl和0.05 M MgCl2。
• 对于DIG标记系统应用：将50 μl NBT溶液和37,5 μl BCIP加至10 ml 0.1 M Tris-HCl中，pH 9.5 (20 °C)，含0.1 M NaCl
• 注：请勿在DIG检测缓冲液中加入MgCl2 ，否则在检测过程后膜上可能会产生斑点背景。碱性磷酸酶不需要Mg2+。
