经过多年的辛苦研发,CST 科学家研发出一款基于质谱的信号通路全景筛查试剂盒 #75676 PTMScan® Multi-Pathway Enrichment Kit 。它自面市以来便享有稳、准、狠的美誉:稳,通路筛查结果可靠;准,允许对一组确定的蛋白质位点以及信号转导节点进行靶向筛选和定量分析;狠,通量极高,且其筛选和定量分析的蛋白质位点和信号转导节点对药物发现、生物标记物发现或诊断学发展至关重要。

该试剂盒主要是由偶联在琼脂糖珠上的抗体及对应的 IP buffer 组成。该试剂盒中的 “Beads” 是实现后续多通路富集实验的核心和关键。这个 “Beads” 的特别之处就是偶联了几千种 CST 高质量的抗体,包含总蛋白的抗体和磷酸化的抗体。既然是用抗体来做实验,那么实验操作就不难。大概的步骤包括样本总肽段制备及纯化—总肽段与抗体孵育的 IP 实验(多通路多肽富集)—富集后多肽的纯化—LC-MS/MS 实验。若需要更详细的实验步骤,可阅读文章 PTMScan® 试剂盒操作方法和关键环节介绍。
试剂盒包含的抗体可富集并定性定量至少 800 种已知的蛋白和至少 4000 个磷酸化位点,涵盖 19 条经典的信号通路,是样本中靶蛋白高通量筛选和检测的稳健方法。有没有一下子明白用这个抗体做一次 IP-MS,相当于做了上千个 WB 呢?
以 PI3 Kinase/Akt Signaling 为例,蓝色高亮的靶点都可被监测,如下图 2

进一步聚类分析的结果如下图(图 3):

运用 PTM Scan® Multi-Pathway Enrichment kit, CST 科学家在人的癌症细胞中通过LC-MS/MS 实验分析了各蛋白分布,最后总结了如下结果图(图4),说明人的癌症细胞中的蛋白磷酸化程度还是很高的,因为激酶的比例占到近1/3(如图4)。该试剂盒提供了对信号通路的综合分析,减少了丢失重要数据点的可能性,可灵活定制后续有针对性的实验。
