关于SHH:
Sonic Hedgehog Homolog (SHH) belongs to a three-protein family called hedgehog. The other two family members are Indian Hedgehog (IHH) and Desert Hedgehog (DHH). Hedgehog proteins are key signaling molecules in embryonic development. SHH is expressed in various embryonic tissues and plays critical roles in regulating the patterning of many systems, such as limbs and brain. SHH also plays an important role in adult, including the division of adult stem cells and the development of certain cancers and other diseases. Human SHH is expressed as a 45kDa precursor, and undergoes a series of processing during secretion. After the removal of the signal peptide, a protease within the C-terminal domain catalyzes the cleavage of SHH into a 20 kDa N-terminal signaling domain (SHH-N) and a 25 kDa C-terminal domain (SHH-C). SHH-N has the “all signaling” capability. SHH-N binds to the 12 pass transmembrane protein Patched (Ptc) on cell surface, which releases the repression of the activity of Smoothened (Smo), a G-protein coupled receptor, by Ptc.

Human Glycoprotein A33 (GPA33) is a single-pass type I membrane protein, belongs to the CTX family of cell adhesion molecular within the immunoglobulin family, can be expressed in normal gastrointestinal epithelium and in 95% of colon cancers. GPA33 consists of one Ig-like C2-type domain and one Ig-like V-type domain. The predicted mature protein includes a single transmembrane domain, a extracellular region and a intracellular tail. Intracellular traffic and recycling to the cell surface appear to play an important role in GPA33 function and to have an influence on its surface density superseding translation regulation. GPA33 has become a promising target of immunologic therapy strategies. GPA33 may also play a important role in cell-cell recognition and signaling.

关于CD55/DAF:
CD55 is a member of the RCA (regulators of complement activation) family. RCA proteins is characterized by the presence of four to 30 SCRs (short consensus repeats also called CCPs for control protein modules) in their plasmaexposed regions. CD55 containing four SCR modules is involved in the regulation of the complement cascade. CD55 is known to bind CD97 via the first SCR. It also binds physiologically generated C3 convertases with its second and third SCRs. Binding results in an accelerated “decay”, or dissociation of active C3 convertases, thus blocking the development of C’ attack complexes on nonforeign cells. It is known that viruses and bacteria also utilize multiple SCR sites for infection.
