文献和实验按照TaKaRa的说明书，我用了Cool Start法，就是所有操作和试剂都是放在冰上进行的，据说能够减少非特异性。我当时担心目的基因量不够，所以P了两管，分到四个孔跑胶回收的。胶回收用的是OMEGA的试剂盒。 切胶回收之后测得目的片段浓度大概为20 ng/ul。按照TaKaRa的pMD18T说明书的要求，pMD18T需要加入1 ul，而目的片段需要0.1 pmol-0.3 pmol，换算一下，需要的目的片段的量大概为0.1*1800*325=58500 pg=58.5 ng到0.3*1800
